The Mercedes Benz kombi was heading towards Kadoma with 23 passengers on board.
Police attributed the accident to speeding.
Acting National Traffic police spokesperson Assistant Inspector Luckmore Chakanza said the driver lost control and the vehicle overturned several times.
One person died on the spot while four others died on admission to Kadoma General Hospital.
Asst Insp Chakanza said 18 people including the driver were admitted at the hospital and treated for injuries.
The driver, he said, is likely to be charged with culpable homicide once investigations have been completed.
Asst Insp Chakanza urged passengers to warn drivers against speeding and report them to police manning roadblocks along the highways.
